دانشگاه اراک سلول و بافت 2228-7035 2 تابستان 90 2011 10 01 Study of Cytotoxic Effects of Taxol and Rosemary Extracts on Cancerous Cells Derived From DMBA-induced Breast Cancer in SD Rats بررسی خواص ضد سرطانی تاکسول و عصاره های الکلی و آبی رزماری بر سلول های سرطان پستان القا شده توسط DMBA در رت های نژاد SD 117 126 958 10.52547/JCT.2.2.117 FA Journal Article 2012 10 29 Aim: The aim of this study was to evaluate the cytotoxic effects of rosemary (Rosmarinus officinalis) extracts on cancerous cells derived from DMBA induced mammary tumor in SD rats. <br />Materials and methods: Treatment group were gavaged by DMBA (10mg/kg) dissolved in 1ml sesame oil. After tumor development, a part of tissue samples were used for pathological studies using H&E staining to determine the type of the tumor. Cancerous cells were prepared after breaking the tissue using mechanical method. Dried leaves of Rosemary were extracted using digestion method with the help of methanol and filtrated by filter paper. One part of extract was kept away, and to separate the non polar compounds, the rest of the extract was washed with non polar solvents (hexane, dichloromethane, ethyl acetate). Then the solvents were removed by rotary evaporator and then the desirable dilutions of extracts as well as Taxol were prepared. After 24 hours treatment of the cells with the extract and taxol the cell viability was evaluated by Trypan blue dye exclusion and MTT colorimetric assay. <br />Results: Alcoholic extract and Taxol showed significant cytotoxic effects on cancerous cells but the effect of aqueous extract was weaker. <br />Conclusion: according to this study alcoholic extract of Rosemary had a dose dependent cytotoxic effect on cancerous cells which was comparable to Taxol cytotoxicity. Moreover these data demonstrated that the induced breast cancer in rat is an appropriate model to simulate human breast cancer. <br />  هدف: مطالعه حاضر به بررسی اثرات سایتوتوکسیک گیاه رزماری (Rosmarinus officinalis) روی سلول‏های سرطان پستان القا شده توسط DMBA در رت‏های نژاد SD پرداخته است.<br /> مواد و روش‏ها: 10 میلی‏گرم از کارسینوژن DMBA به رت های گروه تیمار خورانده شد. پس از ایجاد تومورها، جهت تعیین نوع تومور و بررسی هیستوپاتولوژیک، رنگ آمیزی هماتوکسیلین- ائوزین صورت گرفت. جهت تهیه سلول از روش خرد کردن تومور استفاده شد. از برگ‏های خشک رزماری با استفاده از متانول به روش digestion عصاره گیری شد. سپس عصاره فیلتر شده و مقداری از آن کنار گذاشته و مابقی آن توسط حلال های غیرقطبی هگزان، دی کلرومتان و اتیل استات شستشو داده شد تا مواد غیرقطبی آن جدا شود.  ﻋﻤﻞ ﺣﺬف ﺣـﻼل ﺑـﺎ اﺳـﺘﻔﺎده از ‫دﺳﺘﮕﺎه  تبخیرکننده چرخشی اﻧﺠﺎم ﮔﺮﻓت. از تاکسول و هریک از عصاره‏ها 5 غلظت تهیه شده و  بر روی سلول‏ها اثر داده شد و توانایی زیستی سلول‏ها به دو روش جذب تریپان بلو و رنگ سنجی MTT ارزیابی شد.<br />نتایج: عصاره‏ی الکلی رزماری و تاکسول برروی سلول‏های سرطانی دارای اثرات سمی قابل توجهی بودند ولی عصاره آبی خواص ضدسرطانی بسیار ضعیفی داشت.<br />نتیجه گیری: عصاره الکلی رزمای به صورت وابسته به دز قادر به از بین بردن سلول های سرطانی بوده و خواص سایتوتوکسیک آن قابل مقایسه با داروی تاکسول است. همچنین مدل سرطان پستان القایی یک مدل مناسب برای انجام آزمایشاتی از این دست می‏باشد.<br />  https://jct.araku.ac.ir/article_958_d240e3d38a8882ecad8633c8f9c78c9b.pdf
