دانشگاه اراک سلول و بافت 2228-7035 10 1 2019 03 21 Investigation the Effect of tamoxifen on CTNNBIP1 in cancer stem cells derived from MKN-45 cell line بررسی اثرداروی تاموکسیفن برروی بیان ژنCTNNBIP1 در سلول‏های بنیادی سرطانی مشتق شده از رده‏ی سلولیMKN-45 42 53 36363 10.52547/JCT.10.1.42 FA بهاره قدرتی - دانشگاه رازی، دانشکده علوم، گروه زیست شناسی، کرمانشاه، ایران حسن اکرمی - دانشگاه رازی، دانشکده علوم، گروه زیست شناسی، کرمانشاه، ایران Journal Article 2019 09 07 <strong>Aim:</strong> In the current study, effect of various concentrations of tamoxifen on <em>CTNNBIP1</em> expression gene in the cancer cells derived from <em>MKN-45</em> cell line of gastric cancer was considered as a therapeutic potential. <br /><strong>Materials and method:</strong> The CSCs derived from the <em>MKN-45</em> cell line were treated with different concentrations of tamoxifen for 48 hours and then cell survival was evaluated by the Trypan Blue test. After obtaining IC50, we examined the effect of 100 µM tamoxifen on the expression of <em>CTNNBIP1 </em>in CSCs as a therapeutic potential. <br /><strong>Results:</strong> Analysis of Real-time RT-PCR data showed that 100 μM tamoxifen in a 48-houred treatment can increase the expression of <em>CTNNBIP1</em> gene. <br /><strong>Conclusions:</strong> According to the results, it was concluded that tamoxifen has an inhibitory effect on the Wnt signal pathway and beta-catenin protein by increasing the expression of the <em>CTNNBIP1</em> gene. <br /><strong> </strong> <strong>هدف:</strong> هدف از این تحقیق بررسی اثر غلظت‏های مختلفی از داروی تاموکسیفن بر روی بیان ژن  CTNNBIP1در سلول‌های بنیادی سرطانی مشتق شده از رده‌ی سلولیMKN-45 مربوط به سرطان معده به‌عنوان یک پتانسیل درمانی است.<br /><strong>مواد و روش</strong>‫<strong>ها:</strong> در این تحقیق بقای سلول را با استفاده از تست تریپان بلو ارزیابی شد. پس از به‏دست آوردن IC50 سلول‌های بنیادی سرطانی مورد نظر را با غلظت مشخص دارو طی 48 ساعت تیمار دادیم. با توجه به این اطلاعات به بررسی اثر غلظت‏های مختلفی از داروی تاموکسیفن بر روی بیان ژن  <em>CTNNBIP1</em>در سلول‌های بنیادی سرطانی مشتق شده از رده‌ی سلولیMKN-45 مربوط به سرطان معده به‌عنوان یک پتانسیل درمانی پرداخته شد.<br /><strong>نتایج:</strong> آنالیز داده‌های Real-time RT-PCR نشان داد که غلظت 100 میکرومولار از داروی تاموکسیفن طی تیمار 48 ساعته می‌تواند بیان ژن  <em>CTNNBIP1</em>را افزایش دهد.<br /><strong>نتیجه گیری:</strong> با توجه به نتایج به‏دست آمده می‏توان نتیجه گرفت که داروی تاموکسیفن از طریق افزایش بیان ژن <em>CTNNBIP1</em> بر روی مسیر سیگنالی Wnt و پروتئین بتا کاتنین اثر مهاری دارد.<br /><strong> </strong> https://jct.araku.ac.ir/article_36363_619fbb22733450c579c88736dd7c3198.pdf
