In the third passage, cells divided into 4 different groups and were treated for 1 to 4weeks under the effect of a medium containing extracts of sheep testes as inducer and then expression of sperm maturation genes: Acrosin and Protamine1 were investigated by using of western blotting technique. <br /><strong>Results:</strong> After spermatogonial stem cells treatment by the extracts of sheep testes, variations were seen in cell shape and they convert into sperm- like. Moreover, Acrosin and Protamine1 expression were confirmed. <br /><strong>Conclusion: </strong>examination of induced cells showed that Acrosin and Protamin1 were expressed. Since Acrosin and Protamine1 are the major proteins of the spermiogenesis, it could be concluded that these cells had been completed spermatogenesis stage and started the spermiogenesis stage. <br />  <strong>هدف:</strong> هدف این مطالعه بررسی توان تمایزی سلول‫های بنیادی اسپرماتوگونیال به اسپرم می­باشد.<br /><strong>مواد و روش</strong>‫<strong>ها:</strong> سلول‫های بنیادی اسپرماتوگونیال پس از جداسازی آنزیمی از نمونه­های بیوپسی بیضه افراد آزواسپرمیای غیر انسدادی  در فلاسکT25  کشت داده شدند. در پاساژ سوم سلول‫ها در 4 گروه مختلف به‫مدت 1 الی 4 هفته تحت تاثیر محیط کشت حاوی عصاره­ی بافت بیضه گوسفندی به‫عنوان القا کننده قرار داده شدند و پس از آن بیان  ژن­های بلوغ اسپرم: Acrosin و Protamine1 با استفاده از تکنیک وسترن بلاتینگ بررسی شد.<br /><strong>نتایج:</strong> پس از القای سلول‫های بنیادی اسپرماتوگونیال توسط عصاره­ی بافت بیضه گوسفندی در این سلول‫ها شکل تغییر یافته به­صورت شبه اسپرم در آمدند و  همچنین بیان ژن پروتامین 1و آکروزین تایید شد.<br /><strong>نتیجه­گیری:</strong> بررسی سلول‫های القا شده نشان داد که آکروزین و پروتامین 1 بیان شده اند، از آنجایی‫که آکروزین و پروتامین عمده ترین پروتئین‫های اسپرمیوژنز هستند می­توان این­گونه نتیجه گرفت که این سلول‫ها مرحله­ی اسپرماتوژنز را کامل نموده و وارد مرحله­ی اسپرمیوژنز شده­اند.<br />  https://jct.araku.ac.ir/article_30209_0f47d8af9d0abe580c26d8551f4071f1.pdf
