Investigation of leaf anatomical structure variation among different populations of Marrubium anisodon in Iran

Abstract Aim: In the current study, leaf anatomical structure and tissue cellular variation was evaluated in three populations of M. anisodon in Iran.
Material and Methods: Two individuals were selected from each population. Leaves from middle part of the stems were fixed in F.A.A fixative and thin hand–cut sections were prepared of them. Then leaf samples were double stained and studied using a light microscope. Data were analysed using SPSS and MVSP softwares.
Results: In all studied populations, leaves trans sections were dorsi–ventral. But, qualitative and quantitative traits of leaf differed among the populations. The ANOVA test did not show significant variations among most of the quantitative traits. Populations clustered separately in the UPGMA tree and also PCA and PCO plots. The population’s arrangement pattern did not fit with geographical distances between populations.
Conclusion: Similarity in the ecological conditions of habitats lead to anatomical structural likeness, and this case dose not related with geographical distance. Therefore, wherever the ecological conditions are similar, the likeness of leaf anatomical structure is very possible.
Evaluation of Seed Germination, Growth and Plant Anatomy of Salsola arbuscula Pall. in Salt Stress in In Vitro

Abstract Aim: In this study, the effects of NaCl on seed germination leaf and stem anatomy and growth parameters were investigated in 10 and 60 days-old of Salsola arbuscula species.
Material and Methods: The species seeds were cultivated in WA (0, 100, 200, 250, 300 mM NaCl), and MS (0, 100, 200, 250, 300, 350 mM NaCl) in vitro media. Then 10 and 60 days-old plants were used for studying seed germination and growth parameters. In addition to leaf anatomy of 60 days-old plants in MS (0, 100, 200, 250, 300, 350 mM NaCl) and after recovery in MS (300, 350, 400 mM NaCl), germination percentages were also investigated.
Results: Results showed decline in germination percentage and ability of recovery in germination percentage. Also growth and anatomical parameters showed wide varieties in 10 and 60 days-old plants with increasing NaCl concentration.
Conclusion: S .arbuscula is a halophytic plant that shows a better growth in low concentration of NaCl than control. Also the species protects its water content along the stress condition on the contrary glocophytes and has germinating ability. These characters help to survival of the species.
The Effects of Heavy Metals on Leaf Anatomy, Some Flavonoids and Nitrotoxins of Coronilla varia L. in Hydroponic Culture

Abstract Aim: In this study, the effects of Ni and Zn to leaf anatomy, some flavonoids and nitrotoxin of Coronilla varia were investigated.
Material and Methods: 40 days aged grown Coronilla varia L. plants in equal growth condition were treated with different concentration of ZnCl2 (0, 7.5, 15, 22.5 and 30 mM) and NiCl2 (0, 2.5, 5, 7.5 and 10 mM) for 24 and 72 hours in hydroponic culture and the effects of imposing heavy metals on Leaf anatomy, some flavonoids of control and treated plants examined and compared using 2-DPC (Two Dimensional Paper Chromatography) and TLC (Thin Layer Chromatography) methods. Nitrotoxin measured using spectrophotometer too. All of Data compared together.
Results: Results showed anatomical changes in polluted plants responses to Zn and Ni. In general results showed a wide changes in leaf flavonoids profile with increases number of total flavonoids at Zn and Ni stress. However, Nitrotoxin increased significantly when Zn and Ni increased.
Conclusion: Probably, C. varia L. can tolerate against heavy metals stress of Zn and Ni with changes of flavonoids content and nitrotoxin.
Effect of Cold Stress on the Anatomy and Morphology of the Tolerant and Sensitive Cultivars of Rice During Germination

Abstract Aim: Plants face many stresses in nature and have different defense mechanisms against stress. Rice is a cold sensitive plant and its product considerably decreases under cold stress. Since rice is an important crop, recognizing the tolerant cultivar and their defense mechanism against low temperature seems necessary.
Materials and Methods: In the present study anatomic and morphological features of tolerant (Onda) and sensitive (Nemat) rice varieties were studied at the germination phase under low (10, 15°C) and room temperature (25°C) for a period of of 8 days.
Results: Results show that dry weight, length of the hypocotyl and radicle of both Onda and Nemat cultivars under stress were decreased. The decrease was greater in the Nemat cultivar than the Onda cultivar. In addition, anatomic features of both cultivars under cold stress underwent some changes including increase of thickness of suberin layers and decrease of the amount of aeranchyma tissues.
Conclusion: Changes observed in the two varieties of rice were towards increasing resistance to low temperature stress. The Onda variety had a more suitable defense mechanism compared to the Nemat variety.
