Influence of Occupational Exposures on Sperm Parameters and Chromatin Structure

Abstract Aim: This study examines the sperm parameters (concentration, mobility and morphology) and the state of sperm DNA in infertile men exposed to various occupational exposures.
Material and Methods: This study was performed on 152 men who referred to Isfahan Fertility and Infertility Center )spring 2014 to autumn 2016( for diagnostic purposes. After an interview with all the participants, their semen samples were collected for assessment of sperm parameters and sperm chromatin health, based on the WHO protocol and the TUNEL method, respectively.
Results: The results showed that in men with prolonged sitting, high physiological activity and with exposure to chemical compounds, they had lower sperm motility and higher sperm DNA damage than the presented criteria by previous studies. In addition, in men with DNA damage higher than 10%, there was a significant decrease in sperm parameters.
Conclusion: Occupational exposure has negative effects on sperm parameters and sperm chromatin health, but more studies are needed to prove these results.
Comparison of sperm parameters and its functional characteristics between fertile and infertile men with varicocele

Abstract Aim: The aim of this study is to compare sperm parameters (concentration, motility and morphology), DNA damage, percentage and intensity of ROS (Reactive Oxygen Species) and relationship between these parameters in the fertile and infertile men with varicocele.
Material and Methods: In this study, the semen samples from 83 individuals with varicocele and 32 fertile men were studied. Sperm parameters according WHO (World Health Organization) guidelines, protamine deficiency (chromomycin A3 staining), sperm DNA damage (TUNEL assay), percentage and intensity of ROS (DCFH-DA staining) were assessed in fertile and infertile men with varicocele.
Results: The results of this study showed that sperm concentration, percentage of motility and normal morphology were significantly lower in individuals with varicocele compared to fertile individuals. In addition, percentage of sperm DNA damage, protamine deficiency and ROS positive were significantly higher in infertile men with varicocele.
Conclusion: The increase of stress oxidative and testicular temperature in infertile men with varicocele, lead to decrease the quality of sperm parameters as well as negative effects on sperm chromatin integrity. Therefore the process of spermatogenesis and the rate of fertility were affected in these individuals.
Effects of Different Doses of Silver Nanoparticles on Sperm Parameters, Chromatin Structure and DNA Integrity in Mice

Abstract Aim: The aim of this study was to evaluate the negative effects of different doses of silvernanoparticles on fertility potential, chromatin structureand DNA integrity of epididymal sperm in mice.
Material and Methods: Twenty four male mice were divided into 4 groups (n=6) including one control and three experimental groups for orally administration of silver nanoparticles with three different doses (50, 100 and 200 μl/kg/day) for 5 weeks. Then, cauda epididymal spermatozoa were aspirated for analysis of sperm parameters using general microscopy methods and according to WHO criteria. Chromatin condensation, intensity of sperm chromatin abnormalities and protamine deficiency were assessed by three different cytochemical tests including aniline blue (AB), toluidine blue (TB) and chromomycin A3 (CMA3) tests respectively.
Results: Results showed that third group (treated with the highest silvernanoparticles dose), had the lowest sperm number, the lowest of sperm fast motility percentage and the lowest sperm percentage having normal morphology significantly in comparison with control and others groups. The II and III groups significantly showed differences in sperm chromatin condensation and deficiency comparing to other groups.
Conclusion: In this study the negative effect of silver nanoparticles on mice sperm parameters, sperm chromatin structure and DNA integrity were observed. It is believed that the negative effects of silver nanoparticles on sperm quality are significant and depend on used doses.
Protective Effect of Vitamin E on Sperm Parameters in Adult Rat Treated with Para-nonylphenol

Abstract Aim: The aim of this study was to investigate the effect of vitamin E on sperm parameters in adult rat treated with para-nonylphenol.
Material and methods: Adult male rats were divided into four groups: Control, para-nonylphenol, vitamin E and para-nonylphenl+vitamin E. Oral treatments were performed till 56 days. At the end of treatments, body and left testis weight were recorded and left caudal epididymis was cut in a medium. Released spermatozoa were used to analyze sperm parameters such as sperm number, viability, morphology and motility. Sperm chromatin quality was assessed by nuclear staining using acridine orange and aniline blue.
Results: Body and testis weight as well as normal sperm morphology showed no significant change in four groups. A significant decrease in the number, viability and motility of the sperm was found in rats treated by para-nonylphenol compared to the control. This decrease was significantly compensated by vitamin E in para-nonylphenol+vitamin E group compared to para-nonylphenol group. The application of vitamin E alone could significantly increase sperm viability and motility as comared with the control. Para-nonylphenol had no effect on sperm DNA integrity and histon-protamine replacement compared to the control.
Conclusion: Vitamin E, as a potent antioxidant, could protect the adverse effect of para-nonylphenol on certain sperm parameters in adult rats.
