Total phenol and flavonoid contents of Meristotropis xanthioides Vassilcz. species extract and its protective effect on ethanol-induced hepatotoxicity

Abstract Aim: The aim of this study was to assess the protective effect of Meristotropis xanthioides extract on ethanol-induced hepatotoxicity.
Material and methods: The total phenol and flavonoid contents of the species were measured by Folin Ciocalteu and AlCl3, respectively. Male Wistar rats were divided into three groups: group 1 (control), group 2: received 1ml 50 % ethanol, group 3 : received 1ml ethanol (50 %) plus the extract (500 mg/kg), each group consisted of six rats. All treatments were performed by intragastric administration. Silymarin was used as positive control. Biochemical, histological and morphological analyses were used for evaluation of hepatotoxicity.
Results: The extract had high total phenolic and flavonoid contents. Alcohol consumption significantly increased liver/body weight ratio, amounts of tissue’s malondialdehyde (MDA), H2O2 and also liver enzymes in blood in comparison to other groups. Histological examination showed that alcohol consumption injures liver tissue intensely. All these alcohol-induced changes were effectively inhibited by treatment with the extract.
Conclusion: This research confirmed that the species can represent the protective role on ethanol-induced hepatotoxicity and suggested its capacity to apply as a new healthcare food and drug supplement.
Hepatoprotective effect of hydroethanolic extract of Tragopogongraminifolius in rats induced withCCl4

Abstract Aim: In present study the hepato-protective effects of Tragopogon graminifolius hydro-ethanolic extract (THE) was investigated in an experimental CCl4- induced hepatotoxicity model.
Material and methods: Six groups (n =6) of male Wistar rats were treated with mixture of 2ml/kg BW of sterilized olive oil and CCl4 (1:1), along with 200, 400 and 800 mg/kg BW of THE, respectively. Toxicant group received sterilized olive oil 2ml/kg and CCl4 (1:1) mixture. After 96 hours, liver tissue pathological and serum biochemical parameters (ALT, AST and ALP) studies were done in CCL4+THE different dosages treated groups in comparison with control.
Results: Results showed that receiving CCL4 caused acute inflammation and necrosis in rat liver. THE treated groups significantly showed reduction in all of liver tissue biochemical and histological parameters (p<0.05).
Conclusion: It is concluded that T. graminifolius extract has protective chemical compounds such as antioxidants and flavonoids which are able to protect the liver from damages against CCl4 poisoning by oxidative stresses confrontation.
Study of Hepatoprotective Effect of Malva neglecta L. Hydroethanolic Leaf Extract in Male Rat Induced with Carbon Tetrachloride

Abstract Aim: In this study the hepatoprotectivity effect of Malva neglecta hydroethanolic extract (MHE) were investigated in male rats.
Material and Methods: 35 male rats with 230-250 gr weight were divided in 5 groups: control (taking normal saline,0.5ml/day, IP), sham(taking olive oil, 0.5ml/day,IP),carbon tetrachloride 1:1 with olive oil,0.5ml single dose, IP), treated 1&2 (carbon tetrachloride 1:1 with olive oil,0.5ml single dose and 300mg/Kg and 600mg/Kg MHE /day for 4 days, IP). After the end of examination the blood samples were collected from heart directly and AST, ALT and AP enzymes were analyzed. The microscopic studies of hepatic tissue were done and histological changes observed by (H&E) staining. All data analyzed with ANOVA test. Statistical significance was accepted at p < 0.05.
Results: The serum liver enzymes were increased in rats induced by ccl4 compare to the control group (p < 0.001) and decreased significantly in rats by treated MHE (p < 0.01). Histological finding revealed the necrosis in hepatic tissue. In animals treated with MHE the results showed that the reduction in necrotic cells and increase in hepatocytes regeneration.
Conclusion: Our results showed that the ccl4 has hepatotoxicity effect and it can improve inflammation and necrosis in liver tissue. The MHE have antioxidant and flavonoid that can decrease toxic effect of carbon tetrachloride in liver tissue.
