The Protective Effect of Vitamin E on Spermatogenesis indexes in Rats Treated with Bisphenol A

Abstract Aim: The aim of this study was to investigate the preventive role of vitaminE (Vit.E) on spermatogenesis indexes in rats following exposure to Bisphenol A. Materialand and Methods: Adult male wistar rats with the mean body weight of 231±10g were randomly divided into 4 groups (n=6): control, BPA (250mg/kg/day), Vit.E (150mg/kg/day) and BPA + Vit.E. Oral treatment was performed three times a week till 56 days. At the end of the treatment, the rats were killed and their left testis were weighed, fixed and stained using Heidenhain's Azan methed. Histological and morphometrical analysis of spermatogenesis was carried out. Data were analyzed using One Way ANOVA and the means were considered significantly different at p < strong>Results: Testis weight (p < 0.01) significantly decreased in the BPA group compared to the control. A significant decrease (p < 0.001) in the mean diameter of seminiferous tubules and the germinal epithelium thickness and the indexes of tubular differentiation, spermiogenesis and meiosis were also observed in the testicular tissue of rats treated with BPA compared to the control ones. In the BPA +Vit.E group, the mentioned parameters increased significantly to the control level. Conclusion: Vitamin E can compensate for the undesired effects of BPA on spermatogenesis and therefore could be considered as a therapeutic supplement in the case of BPA toxicity.
Study of the Effect of Bisphenol A on Viability, Morphological Changes and Induction of Apoptosis in Adult Rat Bone Marrow Mesenchymal Stem Cells

Abstract Aim: This study aimed to investigate the effect of Bisphenol A on the cell viability, morphologic changes and induction of apoptosis in bone marrow mesenchymal stem cells of an adult rat. Material and Methods: The bone marrow mesenchymal stem cells were extracted using flashing-out method. At the end of the third passage, cells were divided into 11 groups of control and experimental. Experimental cells were treated with the different doses of Bisphenol A (1, 5, 10, 50, 100, 250, 500, 1000, 2000 and 4000 nM) for four periods of 5, 10, 15 and 21 days in the osteogenic media containing 10% of fetal bovine serum. The cell viability, DNA damage, expression of genes and the morphologic changes of the cells were then investigated during the procedure of osteogenesis. The study data was analyzed using one-way ANOVA and T-Test setting the significant P value at p < strong>Results: Within Bisphenol A treated cells, the mean viability, the mean of nuclei diameter and the expression of anti-apoptotic Bcl-2 gene of the mesenchymal stem cells treated with Bisphenol A significantly decreased (p < 0.05), compared to the control group. In addition the DNA damage and the expression of apoptotic Bax gene of the cells treated with Bisphenol A significantly increased, compared to the control group (p < 0.05). Conclusion: Our data suggest that BPA decreases cell viability and induces apoptosis in mesenchymal stem cells, in a dose dependant manner.
Protective effect of Nigella sativa oil on the Bisphenol A induced-testicular toxicity in adult mice(NMRI): a stereological study

Abstract Aim: The aim of this study was to investigate the protective effect of Nigella sativa oil (NSO),as an antioxidant, on the testicular tissue in mice treated with bisphenol A.
Material and Methods: In this study, 24 adult male mice NMRI were divided into four groups (n=6): control, bisphenol A (200 mg/kg/day), nigella sativa oil (5 ml/kg/day) and bisphenol A + nigella sativa oil. 34 days after oral treatment, the right testis was taken out, fixed and processed followed by Heidenhain's Azan staining. The morphometric parameters of the testicular tissue were evaluated using stereological methods.
Results: The mean volume of seminiferous tubules and it's height of the germinal epithelium, the mean diameter and thickness of the basement membrane, the mean number of spermatocytes, round and long spermatids, spermatogonia and sertoli cells significantly reduced in the bisphenol A group compared to the controls (p < 0.05). The mean volume of seminiferous tubules and the mean number of Sertoli cells significantly increased in the bisphenol A + nigella sativa oil group to the control level, The mean number of spermatocytes and round and long spermatids also showed a significant increase in this group compared to the bisphenol A group (p < 0.05).
Conclusion: Co-administration of nigella sativa oil with bisphenol A can prevent the adverse effects of bisphenol A on the testicular tissue in adult mice.
Stereological study of the protective effect of green tea extract on the testicular tissue of mice treated with sodium arsenite

Abstract Aim: The aim of the present study was to stereological evaluate the protective effect of green tea extract on the morphological changes of seminiferous tubules following sodium arsenite exposure in mice.
Material and Methods: 24 adult male NMRI mice were randomly divided into four groups including control, sodium arsenite (5mg/kg/day), green tea extract (100 mg/kg/day) and finally sodium arsenite+green tea extract. Mice were (orally) treated for 34 days. At the end, mice were sacrificed and their right testis were taken out, fixed, processed and stained with heidenhain' azan method. The morphological and changes of testicular tissue was examined using stereological methods. Data were analyzed using one way ANOVA and means difference was considered significant at p < 0.05.
Results: Sodium arsenite treatment caused a significant decrease in the mean seminiferous tubules diameter (p < 0.008), germinal epithelium height (p < 0.001) and the basement membrane thickness (p < 0.011) as well as in the number of spermatocyte (p < 0.003), round spematid (p < 0.028), long spermatid (p < 0.011) and sertoli (p < 0.03) cells compared to the control group. The above changes increased to the control level in sodium arsenite+green tea exteract group.
Conclusions: The results indicate that green tea extract may be useful in reducing the toxic effects induced by sodium arsenite.
